| Abstract: | The antihypoxant effect of hydroxy- and methoxyphenyl derivatives of quinazolinone-4 have been studied in female rats, including females in the estral cycle stages of diestrus 1/2 and proestrus/estrus, on the model of normobaric hypoxia with hypercapnia. The test substances at a dose of 0.2 of the molecular weight in mg/kg, piracetam and mexidol at a dose of 100 mg/kg, and trimetazidin at a dose of 6.7 mg/kg were administered once intraperitoneally 40 min before testing. Control animals received distilled water. Under the conditions of normobaric hypoxia with hypercapnia, the quinazolin-4(3H)-one derivatives containing a combination of hydroxy- and methoxyphenyl radicals in the structure increased the duration of hypoxia tolerance more significantly than did piracetam and mexidol. Therefore, these derivatives are of interest as potential antihypoxic agents. The antihypoxant effect of the test compounds and piracetam was more pronounced in females at the stage of diestrus 1/2 than in those at the proestrus/estrus stage. Among the hydroxy- and methoxyphenyl derivatives of quinazolinone-4 studied, compound 2-(2-hydroxy-3-methoxyphenyl)-3H-quinazolin-4-one exhibited the most pronounced antihypoxant activity |
